TNF-α对人牙周膜成纤维细胞RANKL/OPG mRNA表达的影响 被引量:9

摘要 目的探讨肿瘤坏死因子-α(TNF-α)对人牙周膜成纤维细胞(HPDLFs)的核因子-κB受体活化因子配体(RANKL)及其饵受体骨保护素(OPG)mRNA表达的影响,了解TNF-α在破骨细胞性骨吸收调节中的作用机制。方法将体外培养的人牙周膜成纤维细胞用不同浓度的TNF-α处理24 h,通过半定量逆转录聚合酶链反应(RT-PCR)来观察人牙周膜成纤维细胞OPG、RANKL mRNA表达强度变化。结果TNF-α上调人牙周膜成纤维细胞的RANKL mRNA表达,RANKL/OPG比率呈上升趋势。结论TNF-α通过调节人牙周膜成纤维细胞的RANKL/OPG的比值可间接地调节破骨细胞分化和活性,从而影响骨代谢。 Objective To study the effects of tumor necrosis factor-alpha(TNF-α)on the gene expression of receptor activator of nuclear factor-κB ligand (RANKL) and osteoprotegerin (OPG) in human periodontal ligament fibroblasts (HPDLFs). Methods HPDLFs were isolated and cultured in media containing 1-100 ng/ml of TNF-α. After 24h incubation, semi-quanitative RT-PCR was performed to detect the expression of OPG and RANKL mRNA using β-actin mRNA as the internal control. Results TNF-α up-regulated the mRNA expression of RANKL, and increased the RANKL/OPG ratio in HPDLFs. Conclusion TNF-α may regulate the differentiation and activities of osteoclasts by enhancing the ratio of RANKL/OPG.
