5-Aza Dc诱导孕酮受体基因启动子脱甲基化可能与DNMT_1表达下调有关

摘要 目的探讨5-杂氮-2′-脱氧胞苷诱导DNA脱甲基化可能机制。方法用5-杂氮-2′-脱氧胞苷处理白血病细胞株HL-60、K562和Jukat,分析处理前后DNA甲基转移酶1 mRNA表达和孕酮受体双启动子(PRA、PRB)甲基化状态的变化。结果5-杂氮-2′-脱氧胞苷处理白血病细胞株后,PRA、PRB都发生脱甲基化作用,DNA甲基转移酶1 mRNA较处理前显著降低(P<0.01)。结论5-杂氮-2′-脱氧胞苷诱导DNA脱甲基化可能与其降低DNA甲基转移酶1 mRNA表达有关。 Objective To discuss the mechanism of 5-Aza-2′-deoxycytidine inducing DNA demethylation. Methods To investigate the changes of DNA methyltransferase 1 (DNMT1) mRNA and methylation status of progesterone receptor double promotors (PRA,PRB) before and after 5-Aza-2′-deoxycytidine treatment. Results PRA and PRB demethylated after 5-Aza-2′-deoxycytidine treatment and DNMT1 mRNA decreased notably comparing with DNMT1 mRNA before treatment. (P〈0.01). Conclusion The demethylation role of 5-Aza-2′-deoxycytidine may be related to the reduction of DNMTI mRNA expression.
