芪术合剂对博莱霉素致大鼠肺纤维化的影响及机制探讨

摘要 目的探讨中药芪术合剂在博莱霉素诱导的实验性大鼠肺纤维化治疗中的作用及其机制。方法将60只健康SD大鼠随机分为正常组、模型组、芪术合剂组和强的松组。经气管滴入博莱霉素复制肺纤维化大鼠模型,并分组予以治疗,于治疗3、7、21d分批处死各组大鼠,每次每组6只。用免疫组化法观察肺组织转化生长因子-β1的表达,并测定透明质酸(HA)含量及支气管肺泡灌洗液炎细胞水平。结果在3、7、21d模型组TGF-β1表达均较正常对照组明显增高(P<0.01),芪术合剂组与同期模型组比较明显降低,芪术合剂3d组高于同期强的松组,而在7d、21d则明显低于强的松组。结论早期运用芪术合剂可抑制TGF-β1和HA的产生,从而预防和阻止实验性大鼠肺纤维化的形成。 Objective To investigate the effect and mechanism of QiZhuHeJi (QZHJ) on bleomyein - induced pulmonary fibrosis in rats. Methods Sixty rats were divided into four groups randomly, including a normal control group, model group treated with bleomyein, QZHJ treated group, and hydrocortisone treated group. The rats were treated by single intratraeheal instillation of bleomyein, and each group was given corresponding drugs respectively. The experimental rats were sacrificed in batch on the 3^rd ,7^th and 21^st day for observing the expression of TGF - β1 and HA, counting the total number of cells inside the BALF and the its differential count. Results The expression ofTGF- β1 in model group on the 3^rd,7^th and 21^st day was obviously increased than that of normal control group ( P 〈 0.01 ). The expression of TGF - β1 in QZHJ group was lower than those of model group, higher than that of hydrocortisone group on the 3^rd day and apparently lower than that of hydrocortisone group. Conclusion The QZHJ has a notable effect in bleomyein induced pulmonary fibrosis rats, inhibition of TGF- β1 expression is one of its possible mechanisms.
