2,3-吲哚醌对人神经母细胞瘤SH-SY5Y细胞增殖影响 被引量:6

STUDY OF THE ANTI-PROLIFERATION EFFECTS OF 2,3-DIOXOINDOLINE ON THE SH-SY5Y CELL LINES

摘要 目的研究2,3-吲哚醌对人神经母细胞瘤SH-SY5Y细胞增殖的影响。方法应用MTT比色法观察2,3-吲哚醌对SH-SY5Y细胞生长的抑制情况;流式细胞仪检测2,3-吲哚醌对SH-SY5Y细胞周期分布的影响;Western blot方法观察其对细胞信号传导通路蛋白有丝分裂原激活的蛋白激酶(ERK)的作用,同时结合免疫细胞化学方法检测2,3-吲哚醌对SH-SY5Y细胞caspase-3的影响。结果2,3-吲哚醌能显著抑制SH-SY5Y细胞增殖,并使细胞周期阻滞于G0/G1期;caspase-3蛋白表达随着药物浓度的升高逐渐增加;加2,3-吲哚醌后磷酸化的有丝分裂原激活的蛋白激酶蛋白(ERK1/2)相对表达量降低。结论2,3-吲哚醌对SH-SY5Y细胞具有抗增殖作用,其作用可能是通过诱导细胞凋亡、改变细胞周期以及作用于细胞通路而实现的。 Objective To study the anti-proliferation effects induced by 2,3-dioxoindoline on SH-SY5Y cells. Methods MTT method was used to examine the effects of 2,3-dioxoindoline on the proliferation of SH-SYSY cells, and flow cytometry to determine the cell cycle. Western blot was employed to assay the change of the key protein, ERK in the cell signal transmission way. The activity of caspase-3 in the SH-SYSY cells was assayed by immunohistochemical technology. Results 2,3-dioxoindoline was found to inhibit the proliferation of SH-SY5Y cells, and disturb the distribution of cell cycle by increasing cell population in G0/Gl phase and decreasing in S phase. The expression of caspase-3 protein was intensified and positively related to the dosage of 2,3-dioxoindoline. And the cell signal transmission way was also affected. Conclusion 2,3-dioxoindoline can inhibit the proliferation of SH-SYSY cells. The mechanism of its anti-proliferation effects is possibly related to the change of cell cycle, the prevention of the signal transmission and the up-regulation of caspase-3 protein in tumor cells.
