正常人眼眶脂肪及眼外肌中抵抗素的表达

Expression of resistin in human orbital adipose tissue and extraocular muscles

目的了解抵抗素在人眼眶脂肪组织及眼外肌组织中mRNA及蛋白表达情况。方法RT-PCR法检测人眼眶脂肪组织及眼外肌组织中抵抗素mRNA的表达情况;免疫组织化学法检测抵抗素蛋白在人眼眶脂肪组织及眼外肌组织中的表达。结果PCR产物电泳结果示抵抗素产物片段约500bp,脂肪组织中含量高于眼外肌组织。抵抗素蛋白表达位于胞浆,免疫组织化学显示人眼眶脂肪组织及眼外肌组织中有抵抗素蛋白存在。结论在人眼眶脂肪组织及眼外肌组织中可检测到抵抗素mRNA及蛋白的表达,抵抗素可能与眼眶炎性疾病,如Graves眼病、炎性假瘤等有一定的关系。

Objective Resistin, an adipocyte derived peptide, has been recognized recently. Resistin is expressed mainly in white adipose tissue,and it is synthesized and secreted by mature adipocytes. Many reports suggested that resistin is an adipokine with potent proinflammatory properties. The objective of this research was to investigate the expression of resistin mRNA and protein in human orbital adipose tissues and extraocular muscles. Methods The orbital adipose tissues （ 12 cases） and extraocular muscles （12 cases） were obtained from the patients of Zhongshan Ophthalmic Center. The expression of resistin mRNA in human orbital adipose tissues and extraocular muscles were detected by RT-PCR. Immunohistochemistry was used to determine the expression of resistin protein. Results There were expression of resistin mRNA in human orbital adipose tissue and extraocular muscles, and the length of PCR products were about 500 bp. The relative gradation of resistin mRNA in orbital adipose tissues was 0. 254 ± 0. 025, and in extraocular muscles was 0. 178 ± 0. 016,showing that the expression of resistin mRNA in adipose tissue was higher than that in extraocular muscles（ P 〈 0.01 ）. Immunohistochemistry showed that the resistin protein was abundant in cytoplasm of adipocytes and myocytes, displaying the brown positive staining in cytoplasm of adipocytes and myocytes. Conclusion Resistin appears to have a close relationship with the orbital inflammatory diseases such as Graves disease, inflammatory pseudotumor, etc.
