摘要
目的:研究手性霉素(Manumycin)联合磷脂酰肌醇-3激酶抑制剂(LY294002)对人卵巢癌细胞株的抑制作用,为临床治疗中晚期妇科肿瘤提供实验资料。方法:Manumycin和LY294002分别及联合在不同药物浓度下作用于人卵巢癌细胞株HO8910PM和OVCAR-3,用MTT法评价其抑癌效果。结果:Manumycin单独使用,对HO8910PM和OVCAR-3的IC50分别为70.3μM/L和59.8μM/L;LY294002单独使用,对HO8910PM和OVCAR-3的IC50分别为76.7μM/L和26.5μM/L;Manumycin与LY294002联用(固定LY294002浓度),对HO8910PM和OVCAR-3的IC50分别为68.5μM/L和32.9μM/L,LY294002与Manumycin联用(固定Manumycin浓度),对HO8910PM和OVCAR-3的IC50分别为65.0μM/L和13.6μM/L。结论:Manumycin和LY294002对人卵巢癌细胞株均有抑制作用,与单独用药相比,二者联用有显著增效作用。
Objective: To study the inhibitory effect of manumyein combined with LY294002, an inhibitor of phosphatidylinositol 3-kinase(PI3K) , on the growth of human ovarian carcinoma cell lines. Methods: Human ovarian carcinoma cell lines HO8910PM and OVCAR-3 were treated with manumyein alone and treated with manumyein combined with LY294002, the proliferation of the carcinoma cells was determined by MTT assay. Results: The IC50 values of HO8910PM and OVCAR-3 treated by manumyein alone were 70.3 μM·L^-1 and 59.8 μM·L^-1, respectively. The IC50 values of HO8910PM and OVCAR-3 treated by LY294002 alone were 76.7μM·L^-1 and 26.5μM·L^-1, respectively. And their ICs0 values treated by manumyein combined with LY294002 (fixed LY294002 concentration )were 68.5μM·L^-1 and 32.9μM·L^-1, respectively. The IC50 values treated by LY294002 combined with manumycin(fixed manumycin concentration) were 65.0μM·L^-1 and 13.6μM·L^-1 , respectively. Conclusion: Manumyein and LY294002 could inhibit growth of Human ovarian carcinoma cell lines. Comparing mamumyein or LY294002 treatment alone, combination of these two components could significantly increase their inhibitory effects on growth of Human ovarian carcinoma cell lines HO8910PM and OVCAR-3.
