摘要
目的体外观察珍珠粉、血竭、乳香、鹿茸、砒石的主要成分(水溶性珍珠粉、血竭素高氯酸盐、乳香水提物、鹿茸多肽、三氧化二砷)对人皮肤成纤维细胞(HSFb)的增殖及胶原蛋白合成的影响,初步探讨其治疗慢性皮肤溃疡的机制。方法组织块法体外培养HSFb;MTT法检测细胞增殖活性;羟脯胺酸比色法测定细胞胶原合成含量。结果药物分别作用于HSFb48h后,水溶性珍珠粉在1.25×10^2~1×10^3mg/L,血竭素在5.0×10^0-2.0×10^1mg/L,乳香水提物在6.4×10^-1~2.0×10^3mg/L,鹿茸多肽在6.25×10^-3~4.0×10^-1mg/L浓度范围内对HSFb具有促增殖作用,与对照组比较差异显著(P〈0.01或P〈0.05),三氧化二砷在1.6×10^-1~8.0×10^-1mg/L浓度范围内对HSn增殖具有抑制作用,与对照组比较差异显著(P〈0.01)。药物分别作用于HSFb48h后,水溶性珍珠粉在2.5×10^2~1×10^3mg/L,血竭素高氯酸盐在2.5×10^0-2.0×10^1mg/L,乳香水提物在6.4×10^-1~2.0×10^3mg/L,鹿茸多肽在1.6×10^-3~4.0×10^-1mg/L浓度范围内对HSFb胶原蛋白合成具有促进作用,与对照组比较差异显著(P〈0.05或P〈0.01),三氧化二砷在3.2×10^-2~8.0×10^-1mg/L浓度范围内对HSFb胶原蛋白合成具有抑制作用,与对照组比较差异显著(P〈0.05或P〈0.01)。结论敛口生肌药珍珠粉(水溶性珍珠粉)、活血生肌药血竭(血竭素高氯酸盐)、活血生肌药乳香(乳香水提物)、温阳生肌药鹿茸(鹿茸多肽)可促进入皮肤成纤维细胞增殖和胶原合成,蚀疮化腐药砒石(三氧化二砷)抑制人皮肤成纤维细胞增殖和胶原合成。
ObjectiveTo observe the effects of main components of pearl powder, dragong blood , boswellin, velver antler and arsenolite ore, including soluble pearl powder (SPD), dracohodin perochlorate (DP), boswellin aqueous extract (BAE), velver antler polypeptide (VAP) and arsenic trioxide (AT), on the proliferation and collagen synthesis of human fibroblasts(HFB) to investigate the therapeutic mechanism on chronic ulcer of skin. Methods We obtained populations of normal human fibroblasts from primary tissue mass culture in vitro. MTT assay was used for observing cell proliferation, and hydroxyproline chromatometry was used for detecting collagen synthesis. Results After HFB was cultured with the tested drugs for 48 hours, the proliferation rate and collagen synthesis of HFB were elevated by SPD, DP, BAE, VAP and lowered by AT. For SPD, DP, BAE and VAP, obvious proliferation -promoting concentrations on HFB were 1.25 ×10^2 -1 ×10^3 mg/L, 5.0 ×10^0-2.0 ×10^1 mg/L, 6.4 ×10^-1 -2.0 ×10^3 mg/L and 6.25 ×10^ -3- 4.0 ×10^ - 1 mg/L, respectively ( P 〈 0.01,P 〈 0.05 ). In concentrations of 1.6 ×10 ^- 1 - 8.0 ×10^-1 mg/L, AT lowered the proliferation rate of HFB. After treating HFB for 48 hours, SPD, DP, BAE and VAP could stimulate collagen synthesis in concentrations of 2.5 ×10^2- 1.0 ×10^3 mg/L, 2.5 ×10^0- 2. 0 ×10^1 mg/L, 6. 4 ×10^-1 -2.0 ×10^3 mg/L and 1.6 ×10^-3-4.0 ×10^-1 mg/L, respectively (P 〈0.05 or P 〈0.01 ), and AT could decrease collagen synthesis in concentrations of 3.2 ×10^-2 - 8.0 ×10^-1 mg/ L. Conclusion SPD, DP, BAE and VAP can elevate,the proliferating rate and collagen synthesis of HFB. On the contrary, AT can decrease the proliferation rate and collagen synthesis of HFB.
