摘要
采用正交实验研究了罗非鱼、鳙鱼、草鱼和鲫鱼4种淡水鱼皮胶原蛋白提取的优化工艺,同时利用eBioService系统分析了纯化胶原蛋白的分子质量和组成。实验结果表明,4种淡水鱼皮胶原蛋白的最佳提取工艺各不相同,在最佳提取条件下,其胶原蛋白提取率依次为36.57%、30.58%、35.14%、45.17%;而得率依次为8.93%、8.52%、9.03%、6.50%。eBio Service系统分析显示4种鱼皮所提取的胶原蛋白电泳区带都与I型胶原蛋白的特征相同,分子质量范围为110~130ku,纯度约为82%~85%。
The paper studied the optimization technology of the extraction of collagen from skin of freshwater fishs including tilapia, bighead carp, grass carp and crucian through orthogonal tests. Furthermore, eBio Service Analysis System was used to investigate the molecular weights and components of the extracted and purified collagen. The results showed that the optimal extracting technology was different among the skins of selected fish. Under the optimal extraction conditions, the extraction ratios of collagens from these skins were 36.57 %, 30.58 96, 35.14 % and 45.17 % respectively, while the corresponding yields were 8.93 %, 8.52 96, 9.03 % and 6.50 % respectively, eBio Service Analysis System showed the SDS- PAGE attributes of the extracted collagens were similar to those of Collagen I, while the molecular weights were 110 - 130 ku with the purity of 82 96 - 85 96.
