温阳活血方对UUO大鼠肾组织TGF-β_1表达的影响 被引量:13

摘要 目的:探讨转化生长因子-β1(TGF-β1)在肾间质纤维化中的表达情况及温阳活血方的干预作用。方法:采用单侧输尿管结扎(UUO)肾间质纤维化模型。将大鼠随机分为6组,即:假手术组、UUO模型组、蒙诺治疗组、温阳活血方治疗组、肉苁蓉治疗组和桃仁治疗组。药物治疗14d后,检测尿β2-微球蛋白(β2-MG),观察肾组织病理改变,并应用免疫组织化学方法检测肾组织TGF-β1的表达。结果:假手术组肾小管上皮细胞、肾间质细胞胞浆TGF-β1有较弱的表达,模型组呈强阳性表达,温阳活血治疗组病理改变较模型组纤维化程度减轻,且TGF-β1的阳性表达较模型组显著减弱,β2-MG排出减少,有统计学差异(P<0.05)。结论:温阳活血方可能通过抑制TGF-β1的表达,从而减少β2-MG的排出,减轻肾小管上皮细胞的损害,保护肾小管功能,减缓间质纤维化的进展。而此方中桃仁的作用优于肉苁蓉。 Objective:To investigate the expression of TGF-β1 in Rats with renalinrerstitial fibrosis and the mechanism protect of Wenyang Huoxie recipe. Methods: Unilateral Ureteral Obstruction rats animal mode was used. The rats were randomly divided into 6 groups: Unilateral ureteral obstruction model group (UUO), Wenyang Huoxie recipe treated group, Monopril treated group, saline cistanche treated group, Semen persicae treated group, Sham - operated group. Renal tissue were examined by light microscopy at the 2th week after treatment. Immunohistoche mistry was applied to measure the expression of TGF -β1 in the renal interstitium. Results. Fibrosis was observed in the renal interstitium of rats with obstructive nephropathy, the expression of TGF-β1 in the normal operation group is low in the renal tubular epithelial cell and renal interstitium cell. Incomparison with SOR group, the expression of TGF-β1 in the of mode group markedly increased (P〈0.05); Incomparison with mode group, Wenyang Huoxie recipe treated group could significantly decrease the expression of TGF-β1 ( P 〈 0.05 ). Conclusion: Wenymag Huoxie recipe can markedly down - regulate the expression of TGF -β1 in the renal tissue of UUO, and reduce the concentration of β2- MG in the urine. Furthemore, Wenyang Huoxie recipe can alleviate the symptom and protect the renal function. Semen persicae is more important than saline cistanche in the recipe.
