不同水系中华绒螯蟹线粒体COⅠ基因片段的RFLP分析 被引量:4

摘要 应用PCR-RFLP技术分析了长江、瓯江和辽河3个水系中华绒螯蟹的线粒体COⅠ的基因片段遗传多样性,应用11种限制性内切酶对COⅠ基因片段进行限制性片段长度多态分析,共检测出6种复合单倍型,其中瓯江群体检测出5种复合单倍型,长江和辽河均有2种复合单倍型。经数理统计分析,3个群体内线粒体DNA的核苷酸多样性指数分别为0.006 7(长江)、0.016 7(瓯江)和0.000 3(辽河),瓯江群体的遗传多态度最高,其次为长江群体,而辽河群体在3个群体中遗传多态度最低。结果说明,中华绒螯蟹具有一定群体内遗传多样性:瓯江群体>长江群体>辽河群体。在群体间,辽河与瓯江群体的净遗传距离最大为0.023 7,长江与瓯江群体间的净遗传距离次之为0.020 7,而长江与辽河群体间的净遗传距离最小为0.004 3,说明3个种群中长江与瓯江种群间的亲缘关系最近,而辽河与瓯江种群间的亲缘关系最远。 The fragments of mitochondrial cytochrome oxidase I of Eriocheir sinensis from Changjiang, Oujiang and Liaohe were amplified by using PCR-RFLP technique. 11 restriction endonucleases were used to digest the PCR prod-ucts. In this study there were 6 composite haplotypes in the 3 populations, there were 5 composite haplotypes in Oujiang population and 2 ones in population of Liaohe and Changjiang. The index of nucleotide diversity (π value) were 0.016 7 (Oujiang), 0.006 7(Changjiang) and 0.000 3 (Liaohe) respectively, indicating that the genetic diversity of Oujiang population was highest among the 3 populations, the genetic diversity of Changjiang population was higher than that ofLiaohe population. Interpopulational net genetic distance was the highest (0. 023 7) between Liaohe and Oujiang, while it was 0.020 7 between Changjiang and Oujiang and was 0.004 3 between Chanjiang and Liaohe samples. The results indicated that the relationship between the populations from Changjiang and Oujiang was the closest, and that of Liaohe and Oujiang populations was the most distant.
