上海地区妊娠高血压综合征患者夫妇HLA-DQB1等位基因频率的研究

摘要 为了探讨HLA与妊娠高血压综合征(PIH)关联的机制,本研究建立了非同位素标记的聚合酶链反应-序列特异性寡核苷酸探针(PCR-SSO)杂交的方法,对PIH病例(患者30例,患者夫妇30对)及89名正常对照进行了HLA-DQB1等位基因型的检测。初步结果显示HLA-DQB1各等位基因型与PIH间无相关性。PCR-SSO方法灵敏度高,可直接检测等位基因,是分析大批量标本的理想方法。本研究为开展大样本的调查及进一步探讨HLA-DQB1各等位基因及HLA-DRB1-DQB1单倍型与PIH的关系打下了基础。 Objective: The objective of our study was to explore the mechanism of association between human leukocyte antigen (HLA) and pregnancy induced hypertension(PIH). Setting: We used polymerase chain reaction sequence specific oligonucleotied (PCR SSO) labeled by nonradioactive method to investigate the HLA DQB1 allelic frequency distribution of PIH patient group (30 patients and 30 couples). 89 Shanghai Chinese were typed as control group. Results: Primary result showed no relationship between HLA DQB1 allelic types and PIH. PCR SSO was an ideal method which may be used to detect alleles directly with high sensitivity especially when sample to be typed was large. Conclusions: Our study has provided a foundation for further study on relationship between HLA DQB1 alleles and HLA DR DQ haplotypes and PIH.
