摘要 庚酸睾酮(TE)诱导的无精子症等同于可逆性的男性不育。为了进一步研究TE对附睾精子成熟的影响,给受试者每周200mg的TE,共注射3个月,分别对注射前,注射后1、2、3个月和恢复期的精浆蛋白质组分进行SDS-聚丙烯酰胺凝胶电泳动态分析。注射TE一个月后精浆中的分子量为87.1、67.6、50.1、39.8和31.6×103(kD)的主要蛋白成分开始下降,分别平均下降到注射TE前的68.0%、40.0%、42.5%、42.3%和30.5%。但注射到精浆中无精子时某些精浆蛋白成分又增加,短暂的增加后又再次降低,甚至到恢复期尚未达到正常的水平,分别平均下降到注射前的42.8%、16.0%、21.8%、29.3%和31.0%。经转移电泳和抗人转铁蛋白免疫化学分析,注射TE后第二个月增加的蛋白成分出现转铁蛋白阳性反应,特别是分子量67.6×103的蛋白成分显著的阳性反应。结果表明精子功能低下与精浆蛋白成分下降时间相吻合。而当精浆中无精子时睾丸支持细胞分泌的转铁蛋白有短暂的增加,可能TE对支持细胞有兴奋作用。 Objectives:To study the mechanism action of testosterone enanathate (TE) on human spermatgenesis. Design: A clinic study on the seminal protein. Settings: National Research Institute for Family Planning. Methods: Eight patients were recruited and injected with TE at a dose of 200mg/week for 3 months. The seminal samples of the patients with TE treatment for 1,2,3 months and recovery phase were subjected to dynamic analysis of seminal protein on the plate of SDS PAGE. Results: The principal protein components (molecular weight:87.1,67.6,50.1,39.8,and 31.6 kD)in the semens of the patients with one month TE treatment began to decrease by 68.0%,40%,42.5%, 42.3% ,30.5%. Some protein components in the azoospermia semen had a transient increase, then decrease again until recovery time. The positive transferrin reaction of the seminal protein component was found after 2 months treatment by the transfered electrophoresis technique,as well as with the immunochemical reaction technique with antibodies against the human transferrin. Especially, protein with a molecular weight 67.6 kD had a significant positive reaction. Conclusions: The study demonstrated that the time when dysfunction of spermatozoa in semen appeared was in accordance with time when the protein component of semen decreased. TE may be able to stimulate the secretory function of Sertoli cells, because the positive transferrin reaction of the protein component in semen was found after 2 months treatment .
