摘要
为探讨室温下己酮可可碱(pentoxifyline,PF)对人不同精子运动特性的影响,将正常精子(I组),少精及运动低下精子(II组)和冷冻复苏精子(II组)分别与PF培育30分钟后洗涤,与对照组同时以计算机精液分析仪(CASA)分析精子运动特性。结果:PF在室温下短时培育能提高各种精子的高活性运动(HA)。其作用在I、I组较明显;PF对平均轨迹速度(VCL)、平均精子头摆动幅度(ALH)及精子运动速度(VAP)的升高作用和对平均直线性比值(LIN)的降低作用经配对t检验,具有显著和极显著意义(P<0.05及P<0.001),且持续至15小时;室温下除LIN以外的精子运动各参数与时间呈负相关,此变化大多于3小时后有统计学差异,以II、II组尤为明显。结果提示:室温下PF可提高精子高活性运动;在人工授精中精子准备时间以选择授精前3小时内进行为宜。
Objective: To observe the effect of pentoxifylline on the movement of human spermatozoa. Study Design: Normal spermatozoa(I), oligazospermia(II) and cryopreserved spermatozoa(III) were exposed to pentoxifylline (1mg/ml) for 30 min. at room temperature(20~25℃). The samples were examined with computer assisted sperm analyser at 0,1,3,6 and 15 hours after treatment with pentoxifylline. Results: The lateral head displacement(ALH), curvilinear velocity(VCL) and path velocity(VAP)were promoted and linearity(LIN) was decreased by exposure to pentoxifylline. When comparing with control groups, these changes were statistically significant (P<0.05 to P<0.001). The movement parameters, except LIN, of the spermatozoa were negatively correlated to the duration after ejaculation. The change was significant after 3 hours at room temperature. Conclusion: Pentoxifylline enhanced the hyperactivation of washed human spermatozoa. Three hours before insemination time should be choosen as the time for sperm preparation for ART.
