猪脾细胞干扰素在细胞中对疱疹病毒的抑制作用

摘要 目的:体外评价猪脾细胞干扰素抗疱疹病毒的活性,为疱疹病毒感染寻找新的治疗手段。方法:主要通过观察细胞病变效应(CPE)、噻唑蓝(MTT)比色法检测细胞存活率和病毒抑制率来评价猪脾细胞干扰素体外对单纯疱疹病毒1型(HSV-1)和2型(HSV-2)的抑制作用。结果:猪脾细胞干扰素对HeLa细胞的半数毒性浓度(TC50)为535.3 IU/ml,对HSV-1和HSV-2的半数有效浓度(IC50)分别为24.6×103IU/L和25.9×103IU/L,治疗指数(TI)为21.8和20.7;其抗病毒效果与人白细胞干扰素及喷昔洛韦一致。结论:猪脾细胞干扰素具有很好的体外抗疱疹病毒作用。 Objective. To evaluate the antiviral activity of pig spleen interferon (psIFN) against herpes simplex viruses (HSV) in vitro. Methods. The antiviral effect of psIFN was studied by assaying infected cells for protection from cytopathic effect (CPE) and by adopting MTT colorimetric assay for viral inhibitory rate. Results. psIFN displayed antiviral activity against HSV-1 and HSV- 2, with 50% inhibitory concentration (IC50) of 24.6 × 10^3 and 25.9 × 10^3IU/L respectively. With the increase of the psIFN dose, the degree of CPE decreased, whereas viral inhibition rate increased. The effect of psIFN on HSV was similar to human interferon (hIFN) and penciclovir. Conclusion. The results indicate that psIFN displayed inhibitive effect on herpes simplex virus in vitro. It could be used as an effective drug for HSV infection.
