摘要
目的:研究Galectin-3在乳腺癌中的表达及其与基质金属蛋白酶MMP-2的表达和微血管密度(MVD)的关系;探讨Galectin-3在乳腺癌发生发展和转移中的可能作用机制。方法:用免疫组化SP法分别检测Galectin-3和MMP-2在84例乳腺癌和32例乳腺良性病变组织中的表达,并用CD34免疫组化染色计数肿瘤血管数量。结果:乳腺癌组织Galectin-3蛋白表达率为71.42%,显著高于乳腺良性病变组织(χ2=11.330,P<0.01)。Galec-tin-3高表达与乳腺癌的分化、分期及淋巴结转移有关。Galectin-3的阳性率在低分化组(92.31%)显著高于高分化(58.33%)、中分化组(64.71%),均为P<0.05。在Ⅲ期和有淋巴结转移组的阳性率显著高于Ⅰ-Ⅱ期和无淋巴结转移组,均为P<0.05。Galectin-3与MMP-2表达呈正相关(P<0.01)。结论:①Galectin-3在乳腺癌中表达增强,它的高表达对判断乳腺癌的恶性程度、浸润甚至转移有重要价值。②MMP-2的高表达使Galectin-3上调在乳腺癌的转移和侵袭中可能起重要作用。
Objective: To study the expression of Galectin-3 and MMP-2 and their relationship with MVD in breast carcinomas. Methods: Expression of Galectin-3 and MMP-2 was detected by immunohistochemistry SP method in 84 breast carcinoma samples, and was compared with that respectively in 32 benign tumors. MVD in breast carcinoma was evaluated by immunohistochemical staining with antibody CD34. Results: Expression of Galectin-3 was detected in a significantly greater proportion in breast carcinoma than in benign tumors (X^2= 11. 330, P〈0.01). The upregulation of the expression of Galectin-3 was related to differentiation and TNM stages (P〈 0.05) and lymph node metastasis (P〈0.05). The positive rate of Galectin-3 in poorly differentiated group was significantly higher than that in well or moderately differentiated group (both P〈 0.05). The positive rate of Galectin-3 in stage Ⅲ and lymph node metastases group was significantly higher than in stage Ⅰ + Ⅱ and none-metastasis groups respectively (both P〈0.05). The expression of Galectin-3 was related to that of MMP-2 in breast cancer (P〈 0.01). Conclusion: ① Expression of Galectin-3 was increased in breast carcinoma. Up-expression of Galectin-3 was useful for evaluating malignancy degree, invasion and metastasis of breast carcinoma. ② Up-expression of MMP-2 increased the expression of Galectin-3 and might play an important role in the metastasis and invasion of breast carcinoma.
