摘要
目的:从雄激素受体表达水平变化的角度,探讨雄激素依赖性前列腺癌向雄激素非依赖性前列腺癌转化的可能机理。方法:体外培养雄激素依赖性前列腺癌细胞株(LNCaP细胞)和雄激素非依赖性前列腺癌细胞株(PC-3细胞),应用流式细胞术测定两种细胞株在生理盐水和二氢睾酮(DHT)作用下雄激素受体(AR)的表达水平。结果:两种细胞株均有AR表达,两种条件下AR的表达水平分别为:LNCaP细胞生理盐水处理组85.99±4.97,DHT处理组93.74±3.63;PC-3细胞生理盐水处理组8.52±1.57,DHT处理组9.52±1.75,两种条件下LNCaP细胞的AR表达水平均高于相应条件下PC-3细胞的AR表达水平,两者有显著性差异(P<0.05)。DHT作用下LNCaP细胞的AR表达水平显著高于生理盐水作用下LNCaP细胞的AR表达水平(P<0.05);DHT作用下PC-3细胞的AR表达水平与生理盐水作用下PC-3细胞的AR表达水平无显著性差异(P>0.05)。结论:雄激素依赖性前列腺癌细胞过表达AR,并且二氢睾酮能促进其表达AR;雄激素非依赖性前列腺癌细胞AR表达较少,二氢睾酮对其AR表达无明显作用,雄激素依赖性前列腺癌向雄激素非依赖性前列腺癌转化可能与前列腺癌雄激素受体表达水平下降有关。
Objective. To analyze the changes of the level of androgen receptor (AR) expression in order to detect the possible mechanism of the hormonal dependent prostatic cancer transform into the hormonal independent prostatic cancer. Methods. Hormonal dependent cell line LNCaP and hormonal independent cell line PC-3 was cultured in vitro. Expression of AR in hormonal dependent cell line LNCaP and hormonal independent cell line PC-3 of prostatic cancer in normal sodium and 5nmol/L dihydrotestosterone (DHT) was quantitated by flow cytometry. Results. AR was expressed in both cell lines, and the level of AR expression of LNCaP was 85.99+4.97 in normal sodium group and 93.74±3.63 in 5 nmol/L DHT treated group, which were respectively higher than that of PC-3 in normal sodium group (8.52 ±1. 57) and in 5nmol/L DHT treated group (9.52±1.75) with significant difference (both P〈0.05). The level of AR expression of LNCaP in DHT treated group was significantly higher than that in normal sodium group (P〈0.05) while the level of AR expression or PC-3 has no difference with that in normal sodium group (P〉0.05). Conclusion: Hormonal dependent prostatic cancer cell overexpressed AR. meanwhile. DHT may promote its xpression of AR. AR scarcely expressed in the hormonal independent prostatic cancer cells and DHT had no influence in its expression or AR. In prostatie carcinoma,the level of AR expression decreased which may be related to the mechanism of the hormonal dependent prostatic cancer transform into the hormonal-independent prostatic cancer.
