鹿瓜多肽注射液对骨折愈合的作用及对血管内皮生长因子表达的影响 被引量:22

摘要 目的:观察鹿瓜多肽注射液经肌肉注射对兔桡骨骨折愈合及愈合过程中血管内皮生长因子(VEGF)表达的影响,并探讨其作用机制。方法:80只新西兰大白兔制成双侧桡骨中段3 mm骨缺损模型,术后随机分为低剂量组、高剂量组、对照组和空白组,分别每日予以0.1 ml/kg鹿瓜多肽注射液、0.2 ml/kg鹿瓜多肽注射液、0.2 ml/kg骨肽注射液或0.2 ml/kg生理盐水。术后1,2,4,6周分批处死动物取材行X线检查、组织学染色、生物力学测试、免疫组化染色及图像分析测定VEGF表达的强度。结果:高剂量组动物骨膜反应出现时间明显早于其他组,骨痂形成的面积也最大;术后6周骨折完全愈合,抗折能力也高于其他组。低剂量组与对照组相比无显著性差异,但均优于空白组。高剂量组和低剂量组VEGF的表达量一直强于对照组和空白组,且表达高峰提前出现,约出现在第2周左右。结论:鹿瓜多肽注射液对兔桡骨骨折愈合有明显促进作用,其有效促进骨折愈合过程中VEGF的合成分泌,是其促进骨折愈合的重要机制。 Objective: To study the effect of cervus and cucumis polypeptide injection on rabbits' radius fracture and to evaluate the effect to the expression level of vascular endothelial growth factor (VEGF), so as to probe into the action mechanism of cervus and cucumis polypeptide injection on fracture healing. Methods: Eighty rabbits were used to establish the models with a 3 mm defect in the middle of radius and divided into four groups randomly as high dose group, low dose group, control group and blank group, which were subjected respectively to intramuscular injection of cervus and cucumis polypeptide injection (0.2 ml/kg or 0.1 ml /kg), bone peptide injection or nothing respectively after operation. Rabbits were sacrificed at the 1st, 2nd, 4th or 6th week postoperatively and extracted the samples, then were examined by radiograph, histology, biomechanical test and immunohistochemistry analysis. Results: The rabbits in high dose group had earlier periosteal reaction and larger bone callus area. After six weeks, the fracture of high dose group had all completely healing and higher strength. There was no significant difference between low dose group and control group, but both groups were better than blank group in healing. The expression of VEGF in high dose group and low dose group was higher than the other groups and its peak value at 2nd week appeared sooner than the other groups. Conclusion: Cervus and cucumis polypeptide injection can promote the healing of rabbit" radius fracture and could actually promote the production and exudation of VEGF through the process of fracture healing.
