摘要
以卷丹百合根尖为试材,对其染色体制片技术的取样时间、预处理液、解离时间、染色剂等方面进行了试验研究,并对3个居群的卷丹百合染色体数目进行了观察。结果表明,根尖取样时间以上午8:30~9:30为佳,用0.1%秋水仙碱溶液4℃下预处理24小时。经卡诺固定液固定6h后,在1mol/L盐酸60℃下恒温水浴中解离8min,改良卡宝品红溶液染色压片镜检,能取得良好的制片效果。卷丹染色体数目观察表明,3个卷丹居群的染色体数目均为2n=3x=36。
The roots of L. lancifolium Thunb. were used to study the effects of the sampling time, the pretreatment chemical solution, hydrolysing time and staining on the chromosome squashing. The results showed that the optimal sampling time of roots was about 8: 30 - 9: 30 am. Among the three pretreatment of chemical solutions, 0.1% colchicine solution got best result. First, 0.1% colchicine solution was pretreated at 4 ℃ for 24 h, then the roottips were fixed with Camoy solution for 6 h, hydrolysed with 1 mol/L HC1 for 8 min at 60 ℃ and stained with modified Carbol fuchsin solution. With this method a very good effect of chromosome squashing could be obtained. The observation indicated that the chromosome number of 3 populations of Lilium lancifolium was 2n = 3x = 36.
