Paxillin在大鼠胰腺不同发育阶段的表达

摘要 目的:探讨与胰岛形成和功能完善相关的基因在大鼠胰腺发育不同阶段的表达趋势.方法:采用高密度寡核苷酸芯片对孕12.5d(E12.5),E15.5,E18.5,新生和成年大鼠胰腺进行基因转录水平分析,并用RT-PCR验证桩蛋白(pxn)和actopaxin在不同发育时期的表达情况.结果:与细胞迁移、聚集黏附相关的基因77.8%在胚胎18.5d高表达,22.2%在新生期高表达;整合素和激酶(ILK)在胚胎18.5d表达量最高,pxn和actopaxin在胚胎15.5d表达量达到高峰;成熟胰腺特异性基因多从胚胎18.5d开始出现明显高表达,调节胰腺细胞分化的相关转录因子表达量多从胚胎18.5d开始下降.结论:Pxn在胚胎发育中后期的高表达可能与胰岛形成及功能完善有关. AIM: To investigate the expression profile of genes related to islet formation and maturation during pancreas development. METHODS: The gene expression patterns in embryonic day 12.5(E12.5), E15.5, El8.5, newbem and adult pancreas were compared by using the GeneChip RAE 230A. The expressions of paxillin and actopaxin at different stages of rat pancreatic development were detected by RT-PCR. RESULTS: Genes related to cell migration and adhesion ( 77.8% ) were expressed at the highest level at El8.5, and the other ones (22.2%) peaked in newbem rats. The expression level of adult pancreas specific genes began to increase from E18. 5, while some transcriptional factors' expression began to decrease from E18.5. Paxillin and actopaxin were expressed at the highest level at E15.5. CONCLUSION: The later embryonic period may be important to pancreas development. Paxillin may play an important role in islet formation.
