摘要
建立了测定氢溴酸加兰他敏含量的高效液相色谱法,采用的色谱柱为十八烷基硅烷键合相,硅胶为填充剂(25 cm×4.6 mm,粒径5μm);流动相为甲醇-乙腈-水(体积比6∶1∶3),流速1 mL/min,紫外检测波长289 nm,柱温为室温,采用外标法测定,进样量20μL。线性范围30-210μg/mL;相关系数为0.999 9;回收率100.1%;相对标准偏差0.65%,与紫外-可见分光光度法比较,该法操作简便,结果准确,专属性好。
A HPLC method for the determination of the content of galanthamine hydrobromide was established. A C18 column (25 cm ×4. 6 mm,5 μm) was used as the stationary phase filled by silica gel;the mobile phase was composed of methanol-acetonitrile-water ( 6: 1 : 3 ) ; at a flow rate of 1.0 mL/min and UV detection wavelength at 289 nm;calculate the content of galanthamine hydrobromide by the external standard; the correlation coefficient of the calibration curve was 0.999 9 with a range of 30 - 210 μg/mL, the average recovery was 100. 1% ;RSD =0. 65%. Compared with the UV absorption spectroscopy,the method is simple, rapid accurate and suitable.
