摘要
目的探讨川芎嗪对大鼠腹膜粘连中TGF-β1表达的影响。方法制作S-D大鼠腹膜粘连模型,关腹前注射川芎嗪注射液(约为50mg/kg)和生理盐水;术后7d行二次开腹手术,链霉亲和素免疫组化法(SABC法)检测粘连组织中TGF-β1蛋白的表达,酶联免疫(ELISA法)定量检测血液及腹腔积液中TGF-β1的浓度。结果川芎嗪组腹腔积液中的TGF-β1浓度(206.37±25.56ng/ml)明显低于模型组(293.42±29.61ng/ml)和生理盐水组(287.46±30.55ng/ml)(P<0.05)。粘连组织中染色强度和血液中的TGF-β1浓度值在模型组(244.98±31.47ng/ml)、生理盐水组(258.14±26.38ng/ml)和川芎嗪组(251.79±27.42ng/ml)三组之间差异无统计学意义(P>0.05)。结论川芎嗪抑制腹腔积液中TGF-β1的表达,但对粘连组织和血液中的TGF-β1含量无影响。
Objective To investigate the role of Ligustrazine in the expression of TGF - β1 in rats with adhesion formation. Methods The uniform defects on the peritoneal wall and cecal surface were developed to establish the rate adhesion model. Ligustrazine(50mg/kg)or 0.6 ml normal saline was instilled into the abdominal cavity. 7 days later,The immunohistochemical method and Enzyme- linked immunosorbent assay was used respectively to identify the volume of TGF - β1 in intraperitoneal adhesion tissue and the concentration in blood or in peritoneal fluid. Results There was no significant difference in the staining density of TGF-β1 in intraperitoneal adhesion tissue and the concentration of TGF-β1 in serum. But the concentration of TGF-β1 in peritoneal fluid was significantly lower in group L rats than those in group M and group N (206. 37 ± 25.56ng/ml versus 293.42 ± 29.61 ng/ml, 258. 14 ± 26.3 8 ng/ml; P〈0.05). Conclusion The Ligustrazine can reduce the expression of TGF-β1 in peritoneal fluid, but not influence those in adhesion tissues or in blood.
