摘要
本实验筛选最强抑酶中药并观察最佳活血中药对肠及胰腺血液动力学的影响。应用体外筛选法观察了九味中药对胰蛋白酶及弹性蛋白酶的抑制作用。各中药提取液按林启寿法进行制备,对胰蛋白酶及弹性蛋白酶活性的抑制作用分别根据张天民及蒋传葵法进行。活血化瘀最佳中药桃仁提取液(HHI-Ⅰ)对肠血液动力学的影响依Leslie法实验,即电磁流量计法测肠血流,应用血氧计测定肠氧耗。进一步的实验中以激光多普勒微循环动态分析仪测定了HHI-Ⅰ对胰组织微循环的影响,并以组织测氧仪观察了胰组织氧分压变化。九味筛选的中药中元胡(Rhizoma Coridalis)对胰蛋白酶及弹性蛋白酶的抑制作用最强,其次分别是大黄、栀子和黄芩、杭芍。犬动物实验结果表明HHI-Ⅰ能改善肠血液动力学,给药后20分钟肠血流增至225.0±68.51ml/min,与给药前(201.34±70.21ml/min)比较差别显著(P＜0.05);给药后40及60分钟进一步增加,分别为245.40±82.78(P＜0.05)和252.20±82.41ml/min(P＜0.01)。给药后肠氧耗也明显增加,60分钟时达5.33±2.57ml/min,与给药前(2.72±1.09ml/min)比较差别显著(P＜0.05)。HHI-Ⅰ还明显改善胰腺微循环及组织氧分压,微循环血流基础值为20.4±5.0ml/min/100g,给药后20分钟增至49.0±9.0ml/min/100g,与基础值比较P＜0.05。
Enxyme-inhibitory effect of some Chinese herbs was screened and pharmacolgic action of the most excellent herb, having blood circulation-improving action, was investigated. Test in vitro was made for screening of the herbs. Dynamic laser Doppler microcirculation analyzer and tissue oxygenmeter were used for research of intestinal and pancreatic hemodynamics. Yuan-Hu(Rhizoma Corydalis, YHI) bad the most potent inhibitory effect on trypsin and e-lestase activity. Tao-Ren(Semen Persicae) injection (HH I - I ) could improve intestinal and pancretic hymodynamics in dog. YHI and HH I - I used together could become the new medicine for treating severe acute pancreatitis.
