摘要
目的:观察乙肝病毒宫内感染新生儿脐血单个核细胞Th1/Th2活化后的功能状态。方法:19例宫内感染新生儿和15例正常新生儿,出生时取脐血抗凝,用梯度离心法分离出单个核细胞,以植物血凝素(PHA)及HBsAg作为刺激物,培养后收获细胞,以CD4-FITC/IFN-γ-PE、IL-4-PE、IgG1-PE标记,FACS流式细胞仪进行测定。结果:PHA刺激下,宫内感染组及正常对照组IFN-γ、IL-4比较,无统计学差异;HBsAg刺激时,两组间IFN-γ有统计学差异(t=2.12,P<0.05);IL-4比较,无统计学意义。结论:在HBsAg刺激下,IFN-γ表达不足,提示宫内感染组存在特异性免疫耐受,Th1水平的提高在控制乙肝病毒感染中起重要作用。
Objective: To observe functional status of activated mononuclear cell T Helper Cells 1, 2 of umbilical blood with intrauterine infection of Hepatitis B virus. Methods: The cord blood of the 19 cases neonate with intrauterine infection and 15 cases normal newborn (study group) were anticoagulated when they were born. Mononuclear cell was isolated by gradient centrifugation and stimulated by phytohemagglutinin (PHA) and HBsAg, harvesting after cultivation , tag with CD - FITC/IFN - r - PE, IL -4PE, IgG1 - PE, determine by FACS flow cytometry. Results: Under stimulation of PHA , there had no difference of IFN - r, IL - 4 between group with intrauterine infection and control group; When excited by HBsAg , there had difference of IFN - r (t = 2. 12, P 〈 0. 05 ) , but no statistical significance of IL -4 in two groups. Conclusion: Under stimulation of HBsAg, IFN -r express insufficient , which indicate there has specific immunologic tolerance in intrauterine infection and elevation of TH1 level has important effection in controlling HBV infection.
