摘要
目的研究干细胞因子(SCF)和碱性成纤维细胞生长因子(bFGF)在体外对间充质干细胞(MSC)的作用特点,探索体外大量培养间充质干细胞的方法和条件。方法用梯度离心和贴壁培养法分离MSC,进行表面分化抗原的鉴定。培养体系中分别加入SCF、bFGF或SCF+bFGF,用MTT实验和检测细胞周期来观察细胞的增殖情况。结果加入SCF和bFGF后MSC的增殖活性增强,两种因子作用强度相近,两者合用刺激增殖的效果未见增强。结论SCF和bFGF有刺激MSC增殖的作用,可以在一定程度上延迟MSC在体外培养过程中的退化现象。
Objective To study the effects of stem cell factor(SCF) and basic fibroblast growth factor(BFGF) on mesenchymal stem cell(MSC) in vitro. Methods Gradient centrifugation and adherent culture were used to get a great amount of NSC and then differentiation antigens were identified. SCF, BFGF or SCF + BFGF were added to the medium, and then, MTT test and the test of cell cycle were made to study cell proliferation. Results Both SCF and BFGF could enhance the proliferation with similar results. The combined use of SCF and BFGF produced no better results. Conclusion SCF and BFGF can enhance proliferation and therefore, can delay the regression of MSC in vitro to some degree.
