摘要
目的探讨促排卵药对小鼠子宫内膜发育的影响,为临床用药提供实验依据。方法随机将160只雌性昆明小鼠分为非孕组和受孕组,每组又分为对照组、克罗米酚组(CC组)、单纯促排卵组(HMG+HCG组)和GnRHa+单纯促排卵组(GnRHa+HMG+HCG组)4个小组,每小组20只。各用药组分别给予相应的促排卵药,对照组给予生理盐水。于用药后24 h和48 h分批处死非孕组小鼠,取子宫做成石蜡切片,HE染色,观察子宫内膜发育情况。受孕组的分组和用药与非孕组完全相同,于非孕组雌鼠处死取材的当天与雄鼠合笼,让雌鼠受孕。于受孕后6.5 d处死孕鼠,观察孕鼠着床情况以及胚胎发育指标。结果非孕组:对照组子宫内膜厚度明显厚于3个用药组(P<0.05),3个用药组中,GnRHa+单纯促排卵组子宫内膜厚度以及腺体发育略优于另外两组,但无显著性差异(P>0.05)。受孕组:对照组胚胎着床率明显高于用药组(P<0.05),各用药组之间有差异,但无统计学意义(P>0.05)。结论在三种促排卵方案中,GnRHa+单纯促排卵方案对子宫内膜的影响最小,内膜与间质发育的同步性最好,着床率较高。
Objective To explore the effects of ovulation simulants on mouse endometrium and embryon nidation and development so as to provide experimental basis for clinical drug administration. Methods 160 Kunming female mice were divided into the non-pregnancy group and the pregnancy group. Each group was further divided into four subgroups: the contrast group, the clomiphene citrate group (CC group), the simple human menopausal gonadotropin group (HMG group) and the human chorionic gonadoiropin group (HCG group), and the combined group consisting of the HMG group, the HCG group and the tropin-releasing hormone group(GnRH group). Each subgroup consisted of 20 mice. The contrast group was given normal saline injections, while all other experimental groups were given different ovulation-stimulation drugs. Mice in the non-pregnancy group were executed in 24 hours and 48 hours after the drug administration to remove their uteri which were then made into paraffin sections for observation of endometrial development. Mice in the pregnancy group were executed on the 6.5th day afrer their pregancy to observe their embryonic nidation and development. Results In the non-pregnancy group, endometria in the contrast group were found thicker than those in the other three groups( P 〈0.05). The endometria were thicker and the glandular development was better in the combined group than the other two drug-administration groups, but no significant differences were found ( P 〉 0.05). In pregnancy group, the embryonic development in different subgroups showed no significant differences ( P 〉 0.05). Conclusion The combined group had the least effects on endometria, and showed the best synchronizing characteristics of intimal and interstitial development and high rate of embryonic nidation.
