大孔树脂法纯化泽泻总萜醇类成分的实验研究

Experimental Study on Enrichment and Purification of Total Terpenoid Components from Alisma Orientalis with Macroporous Resin

探讨用大孔树脂富集纯化泽泻总萜醇类成分的方法。以乙酰泽泻醇B为对照品,用分光光度计测定泽泻提取物及不同树脂洗脱部位的总萜醇类成分含量。结果:以AB-8型树脂的富集纯化性能最佳,最优组合:半径、柱高比为1∶10;药液浓缩比为1∶1;药液上样与洗脱速度为10 ml/min;药材与树脂的用量比为4.6∶1;洗脱溶媒为70%乙醇;洗脱液量为4倍床体积。结论:AB-8大孔树脂可有效用于泽泻总萜醇类成分的富集与纯化。

To discover the method of enrichment and purification of total terpenoid components from A lisma Orientalis with macroporous resin. Spectrophotometer was used for detecting the contents of total terpenoid components in the extracts of Alisma and various parts of elution with different kinds of macroporous resin, compared with acetylalisol B. The results showed that the resin AB-8 is the best in character of enrichment and purification of total terpenoid components from A lisma Orientalis. The optimal parameters were radius： height 1：10, the ratio of condensing 1：1, the speed of loading and eluting 10 ml/min, the amount ratio of raw herb and resin 4.6：1, 70% alcohol as the elution solvent, the elution volume 4BV. Conclusion： The macroporous resin AB-8 could be effectively used to enrish and purify the total terpenoid components from Alisma Orientalis.
