中国野桑蚕mtDNA中Cyt b基因的克隆及序列分析 被引量:1

摘要 通过PCR的方法,对中国青州野桑蚕mtDNA中Cytb基因及侧翼序列进行克隆和序列分析。所获得的序列长度为1761nt,含有完整的Cytb基因t、RNA-ser基因及ND6和ND1基因的部分序列。同源性分析显示,所测定的Cytb基因与中国安康野桑蚕、日本野桑蚕、中系夏芳家蚕、中系C-108家蚕、日系Aojuku家蚕和韩国Backokjam家蚕的Cytb基因具有很高的同源性;基于线粒体Cytb基因的核苷酸序列的分子系统进化分析显示,中国青州野桑蚕与家蚕的亲缘关系较近,表明家蚕可能起源于中国野桑蚕。 The Cyt b gene and its flank region (1761nt) of mitochondrial DNA (mtDNA) from the Chinese Qingzhou Bombyx mandarina were cloned using PCR. Sequence result revealed the fragment contained an intact Cyt b gene, tRNA-ser (UCN) gene, and partial sequence of ND6 and ND1 gene. Homologous comparison of Cyt b genes from Chinese Ankong Bombyx mandarina with Japanese Bombyx. mandarina, Chinese Bombyx mori (strain Xiafang) , Chinese Bombyx mori (strain C108), Japanese Bombyx mori (strain Aojuku) and Korean Bombyx mori respectively showed high homology. Molecular phylogenetic analysis based on the sequence of Cyt b gene reflected the relationship between Qingzhou Bombyx mandarina and Bombyx mori was closer, it indicated that Bombyx mori possibly derivated from Chinese Bombyx mandarina.
