摘要
目的建立同时测定蜂蜜中槲皮素和山奈酚含量的方法。方法以Hpersil BDS C18(250 mm×4.6 mm,5μm)为分析柱,流动相为甲醇+0.4%磷酸水溶液(45+55)并用三乙胺调节pH至4.2,流速为1.0 ml/min,检测波长为375 nm。结果槲皮素的线性范围为0.045-2.25μg,山奈酚的线性范围为0.052-2.600μg,相关系数均为0.999 1;平均回收率分别为95.9%,97.6%。5种蜂蜜中均含微量槲皮素(18.62-189.26μg/g)和山奈酚(5.97-22.54μg/g),其含量与蜜源有关,桉树蜜中槲皮素含量最高(189.26μg/g),红树林蜜中山奈酚含量最大(22.54μg/g)。结论该方法简便、准确,可以用于同时测定蜂蜜中槲皮素和山奈酚的含量。
Objective To establish a method for the determination of quercetin and kaempferd in five kinds of honeys simultaneously.Method The determination was carried out with Hpersil BDS C_(18)(250 mm×4.6 mm,5 μm) column.The mobile phase consisted of methanol+0.4% phosphonic acid(45+55) and pH was adjusted to 4.2 by triethylamine.The flow rate was 1.0(ml/min).The detective wavelength was 375 nm.Results The standard curves of quercetin and kaempferd are linear within the range of 0.045~2.25 μg,0.052~2.600 μg and the coefficient are(0.999?1,)(0.999?1).The reclamation are 95.9%,97.6%.The content s of both quercetin(18.62~189.26(μg/g)) and kaempferol(5.97~22.54(μg/g)) are lower in the five kinds of honey.The contents of quercetin and kaempferol had close relation with the plant that honey came from.The eucalyptus honey has the highest contents of quercetin(189.26(μg/g)),the mangroves honey had the highest contents of kaempferol(22.54(μg/g).) Conclusion The method is simple and accurate and can be used to determine the contents of quercetin and kaempferol in honey simeltaneously.
