摘要
目的研究奈替米星对于豚鼠耳蜗毛细胞的凋亡作用。方法将豚鼠分为生理盐水对照组和奈替米星组,连续给药14 d,利用TUNEL法原位检测动物耳蜗毛细胞的凋亡情况。结果生理盐水对照组豚鼠耳蜗未见凋亡毛细胞,而奈替米星组豚鼠耳蜗基底回中第1、2排外毛细胞丢失,第3排外毛细胞呈阳性表达,第3回中第1排外毛细胞丢失,第2排外毛细胞呈阳性表达。第2回毛细胞中仅见第1排外毛细胞呈阳性表达。结论奈替米星作用于豚鼠耳蜗时可引起外毛细胞凋亡;奈替米星耳毒性的产生可能与其引起耳蜗毛细胞凋亡有关。
AIM To study the effect of netilmicin on apoptosis of guinea pig cochlear hair cells. METHODS Twenty-four guinea pigs were randomized to normal saline (NS) group and netilmicin group. After 14 days' administration consecutively, apoptosis of their cochlear hair cells was detected by TUNEL method in situs. RESULTS In NS group, apoptosis of hair cells was absent,but in netilmicin group the disappearance of outer hair cells(OHCs)was observed both in the first and second row of the cochlear basal turn and in the first row of the third turn, labeled nuclei can be seen in OHCs in beth the third row of basal turn and the second row of the third turn, as well in OHCs in the first row of the second turn. CONCLUSION Apoptosis of cochlear OHCs can be induced by netilmicin. Ototoxicity induced by netilmicin maybe related to apoptosis of cochlear OHCs.
