中药861合剂对体外培养贮脂细胞Ⅳ型胶原mRNA的影响

THE EFFECT OF HERBAL COMPOUND 861 ON TYPE N COLLAGEN mRNA LEVEL IN CULTURED RAT LIPOCYTES

目的观察８６１合剂对体外培养贮脂细胞Ⅳ型胶原ｍＲＮＡ的影响。方法与结向传一代的雄性Ｗｉｓｔａｒ大鼠肝脏贮脂细胞培养液中加入终浓度为１０ｍｇ／ｍｌ的中药８６１合剂（０．４５μｍ过滤）或０．６２５ｍｍｏｌ／Ｌ的秋水仙素，另设空白对照组，继续培养４８小时，收获贮脂细胞提取总ＲＮＡ，以地高辛配基标记的小鼠Ⅳ型胶原ｃＤＮＡ探针与之做斑点杂交，以β－ａｃｔｉｎ作内对照，结果显示８６１合剂组及秋水仙素组贮脂细胞Ⅳ胶原ｍＲＮＡ水平较空白对照组明显降低，而且８６１组比秋水仙素组降低更明显。结论认为８６１合剂的抗纤维化机理之一是调控贮脂细胞的Ⅳ型胶原表达。

To explore the in vitro effect of herbal compound 861 on type Ⅳ collagen mRNA levels in rat lipocytes. Methods The second passage of lipocytes was exposed to 861 at a final concentration of 10mg/ml, and another group was exposed to colchicine at a concentration of 0.625mmol/L. After 48 hours, the lipocytes were harnested and total RNA was isolated to detrmine type Ⅳcollagen mRNA by dot blot hybridization with digoxigenin labelled cDNA probes. Results The results displayed that type Ⅳ mRNA collagen level in 861 and colchicine treated lipocytes remarkably decreased than that in blank control. Furthermore, the decrease of type Ⅳ collagen mRNA level in 861 treated lipocytes was even more profound than that in colchicine treated lipocytes. Conclusions The anti-fibrotic effect of herbal compound 861 is probably achieved by modulating type Ⅳ collagen expression in lipocytes.
