内蒙古西部地区砂珍棘豆(Oxytropis racemosa Turcz.)遗传多样性分析 被引量:5

摘要 用ISSR分子标记对内蒙古西部地区砂珍棘豆(Oxytropis racemosa Turcz)3个不同生境的地理种群进行了种群遗传多样性分析.结果表明砂珍棘豆种群具有较高的遗传多样性,14个ISSR引物扩增出284条带,Shannon多样性指数I=0.3630,Nei′s多样性指数H=0.2234,物种总基因多样性(HT)为0.2236,种群内基因多样性(Hs)为0.2085,大部分遗传变异(93.27%)存在于种群内,6.73%的遗传变异存在于种群间.遗传分化系数(Gst)为0.0673,基因流(Nm)为6.9318,说明种群间有很强的基因交流,遗传漂变不会引起遗传分化.UPGMA遗传距离聚类结果表明,3个地理种群中,乌审旗和伊金霍洛种群先聚在一起,然后与鄂托克种群聚在一起,遗传距离与地理距离相关, The genetic diversity of Oxytropis racemosa Turcz. was analyzed using inter simple sequence repeat (ISSR) markers. The results indicated that there was relatively high genetic diversity in O. racemosa Turcz in Inner Mongolia western regions. 284 bands were amplified from 3 populations in different habitats with 14 reliable primers. The Shannon information index I was 0. 3630, and Nei's index H was 0. 2234. The total gene diversity Ht was 0. 2236, and the gene diversity within populations Hs was 0. 2085. 93.27% of genetic variation existed within populations while 6.73% of genetic variation existed among populations. The Gst was 0. 0673,and the gene flow was 6. 9318. There was a strong gene flow among the populations. So the genetic drift would not cause the genetic differentiation. The UPGMA Dendrogram displayed that the Wushen and Yijinhuoluo populations clustered first, and then they clustered with the Etoke population. The results showed the genetic distance related to geographic distance in three populations of Oxytropis racemosa Turcz.
