摘要
本实验采用纯系SD雌性大鼠为实验动物,在背下部伤口上分别于手术后第1,4天,第4,7天,第7,10天和第10,13天,共4组行X线照射,每次照射6GY,两次共12GY,并分别于手术后第7,10,20,40天取材,在光镜下观察伤口胶原形态学变化。以IBAS-2000(Opton)计算机图像分析系统分析胶原密度变化。光镜下观察:胶原纤维排列紊乱,并较细;照射组的胶原平均光密度明显小于对照组。本实验研究提示:通过连续X线照射后伤口皮肤胶原纤维合成和代谢的抑制作用以手术后第7天内影响最大,第7~13天的作用也明显高于对照组。通过本实验研究提出了瘢痕疙瘩、增生性瘢痕切除后,在行放射线照射时应以第4,7天为中心。
WeinvestigatedthehistologicalstudyonX-Rayafectingcolagenousfibersduringwoundhealing.WeusedinbredoinefemaleSDratsaslaboratoryanimals.Theyweredividedintofivegroups:fourgroupsunderwentirradiationonthelowerback(irradiatedgroups)respectivelyon1stand4thday,4thand7thday,7thand10thday,10thand13thdayaftertheoperation,andthefifthgroupbeginthecontrolgroup.Theexposuredosewas6GYeachtime.Tissuesweretakenatthewoundarearespectivelyon7thday,10thday,20thdayand40thdayoftheoperation.Weusedlightmicroscopestostudymorphologicalchangesonthesetissues.IBAS-2000Computerizedimagepatternanalysissystem(OPTON)wasusedtoanalysethechangesonthedensityofcolagenousfibers.Onhtelightmicroscopeexamination,colangenousfiberswereirregularandslendered,tissuesfromtheirradiatedgroupsre-vealedadecreasedofthecolagenousfibersdensity.Studyingthehistologicalchangesofthewoundafterfractionatedirradiation,weconcludedthatin7thdayaftertheoperation,X-Rayhasitsmaximalinhibit-ingefectoncolagenousfiberssynthesisandmetabolism.Theefecton7thand10thday10thand13thdayirradiatedgrougsisalsogreaterthanthatofthecontrolgroup.Wesuggestthatincasesofkeloidorhypertrophicscar,Wheneveriradiationtherapyisconsidered,theprotocolshouldbecenteredon4thand7thdayaftertheresection,Wepointedouttheimportanceofanearlierirradiation.
