摘要 采用pH-stat法探讨了Alcalase 2.4L FG酶水解赤豆蛋白工艺.应用二次回归通用旋转正交设计和SAS软件,就主要工艺参数对水解度的影响进行响应面分析,建立了相应数学模型,并采用脊岭分析,优化了工艺参数;探讨了该酶催化水解赤豆蛋白反应的动力学参数:Km=0.0969 mol.L-1,Vmax=0.00307 mol.min-1.L-1. The kinetics of hydrolysis on phaseolus angularis protein with alcalase 2.4L FG by pH - stat method was investigated:the hydrolysis condition was discussed and optimized by response surface analysis and ridge of maximum response, respectively, in SAS software by second order regression rotation designing. The macimum velocity and instant K were determined through probing into the relation of the substrate concentration and reaction velocity : Km=0.0969mol·L^-1,Vmax=0.00307mol·min^-1·L^-1.
