全反式维甲酸体外诱导神经母细胞瘤细胞分化的实验研究 被引量:6

摘要 目的体外观察全反式维甲酸(all trans retinoic acid,ATRA)对神经母细胞瘤(neuroblastoma,NB)细胞系SH-SY5Y(SY5Y)细胞生长及分化的影响及其可能的分子机制。方法用RT-PCR方法检测ATRA作用前后SY5Y细胞TrKA mRNA表达情况;采用台盼蓝拒染法检测ATRA对SY5Y细胞的体外抗增殖作用;用倒置相差显微镜观察细胞形态变化。结果ATRA作用7d后SY5Y细胞TrKA mRNA表达增高;ATRA对SY5Y细胞有抗增殖作用和诱导细胞分化作用,并呈一定时间和剂量依赖关系。结论ATRA对SY5Y细胞有生长抑制及诱导分化作用,该作用与上调SY5Y细胞TrKA mRNA表达水平有关。 [Objective] To study the effect of ATRA on growth and differentiation of SH-SY5Y cell line and possible molecular mechanisms. [Methods] The expression of TrkA mRNA was detected by RT-PCR. Trypan blue exclusion was used to detect the antiproliferative effect of ATRA on SY5Y cells. Morphologic changes were observed by using phase-contrast microscopy. [Results] TrkA mRNA was upregulated by ATRA. ATRA could induce morphological differentiation and inhibit growth of SY5Y cells and showed time and dose dependent manner. [Conclusion] ATRA induced morphological differentiation and inhibited cells growth in SY5Y cell hne and this may be related to upregulation of TrkA mRNA.
