Negri小体内的狂犬病毒抗原

摘要 以含Negri小体(NB)的7例海鸟及4例小脑切片,分别作Macchiavello(马氏)和磷钨酸伊红(EPT)染NB,以单克隆-狂犬病毒核衣壳蛋白抗体为一抗作SP染色,并作免疫加乌氏和/或EPT的双重染色。见NB内部含狂犬病毒核衣壳蛋白抗原(RVAg)较少,免疫反应较弱,只外围仍见免疫反应较强之外环。少数NB内有内小体,也由免疫反应强阳性之小环及阴性之内容物构成。在小脑可见免疫反应极弱甚至阴性之NB。这些现象提示NB的形成可能是狂犬病毒体(RV)的退变现象。 From 7 cases of human rabies containing Negri body (NB) in brain, 7 blocks of hippocampus and 4 blocks of cerebellum were sectioned and stained with (1) Macchiavello's method and eosin phosphotungstic acid (EPT) staining to demonstrate NB, (2) immune staining with monoclonal anti-rabies nucleocapsid protein antibody,and (3) double staining:immune staining followed by Macchiavello or ETP.In comparison with non-NB rabies virus antigen (RVAg) particles, the amount of RVAg inside NB was less, while the outer rim of NB remained strongly positive. Inner bodies of NB were found in 3 cases, and they were also composed of strongly positive outer rim and negative inner content. In cerebellum, faintly or negatively stained NBs were found inside Purkinje's cells.These findings indicate that the occurrence of NB is probably due to some regressive changes of rabies virus in these areas.
