Expression of Hypoxia Inducible Factor-1α in Gastric Carcinoma and its Relationship with Vascular Endothelial Growth Factor

摘要 [目的]探讨胃癌组织中缺氧诱导因子-1α(HIF-1α)的表达及其与血管内皮生长因子(VEGF)的关系。[方法]用免疫组织化学SP法检测80例胃癌中HIF-1α、VEGF的表达,并与30例胃癌旁不典型增生组织、30例胃正常组织作为对照;分析HIF-1α、VEGF与胃癌临床病理因素及预后的关系。[结果]HIF-1α和VEGF在胃癌中的阳性表达分别为47.5%(38/80)和71.3%(57/80),明显高于胃癌旁不典型组织及胃正常组织(P<0.05)。HIF-1α、VEGF与胃癌的TNM分期以及肿瘤浸润深度、淋巴结转移等相关(P<0.05),与组织学分级无关。胃癌中HIF-1α与VEGF的表达呈显著性正相关(r=0.328,P<0.01)。[结论]HIF-1α、VEGF均在胃癌中过表达并且与胃癌的侵袭、转移及预后密切相关,HIF-1α可能通过上调VEGF的表达,在胃癌的发生、发展中发挥重要作用。 [Purpose] To explore the expression of hypoxia inducible factor-1α(HIF-1α) in gastric carcinoma and its relationship with vascular endothelial growth factor (VEGF). [Methods ] The expression of HIF-1α and VEGF in 80 cases with gastric carcinoma, 30 cases with gastric paracarcinous tissue (mucosal atypical hyperplasia tissue) and 30 cases with normal gastric tissue(control groups) were detected by immunohistochemistry technique (SP). The relationship between the expression of HIF-1α, VEGF and clinicopathologic feature, prognosis of gastric carcinoma was analyzed. [Results] The positive expression rates of HIF-1α and VEGF in gastric carcinoma were 47.5%(38/80) and 71.3%(57/80) respectively, and both significantly higher than those in gastric paracarcinous tissue(mucosal atypical hyperplasia tissue) and the normal gastric tissue (P〈0.05). The expression of HIF-lotand VEGF in gastric carcinoma correlated to TNM stage, invasive depth and lymph-node metastasis (P〈0.05), not to histological grade. There was significantly correlation between HIF-1α and VEGF expression(γ=0.328,P〈0.01). [Conclusion] The over-expression of HIF-1α and VEGF in gastric carcinoma closely relates to invasion, metastasis and prognosis in gastric carcinoma. HIF-1α may play an important role in carcinogenesis of gastric carcinoma by up-reaulate the expression of VEGF.
