摘要
为阐明内毒素休克时机体免疫功能的改变提供新的思路,用未成年小鼠观察内毒素对胸腺细胞凋亡的影响。结果表明,内毒素可引起小鼠胸腺重量、胸腺细胞总数和活细胞总数明显减少,胸腺死亡细胞百分率和DNA片断百分率均明显增加,且呈正相关,DNA电泳示胸腺细胞出现典型的"ladder"(梯状)带型,电镜检查可见细胞凋亡特有的形态学变化,流式细胞仪分析示胸腺细胞凋亡百分率明显增加。提示内毒素可引起小鼠胸腺细胞凋亡,其后果与内毒素休克时机体免疫力降低可能有关。
To illustrate the immune change of body in endotoxic shock, thymic apoptosis of immature mice induced by endotoxin is observed. Endotoxin results in decrease of thymic weight, total cell number and viable cell number. It can cause increase of both dead cells and NDA fragments. The electrophoresis shows ladder-like shape of DNA. Unique features of apoptosis could be seen by electron microscope. The flowcytometry shows an increased percentage of apoptosis. The result is endotoxin can induce thymic apoptosis of mice, which may due to decreased immunity of endotoxic shock.
