OXIDATIVE STRESS INDUCED APOPTOSIS IN HUMAN PERIPHERAL BLOOD LYMPHOCYTES

摘要 为了证明氧自由基诱导细胞凋亡,以正常人外周血淋巴细胞为研究对象,在体外培养条件下,用H2O2处理后,从形态及生物化学方法观察凋亡现象。H2O2处理8h后,形态上出现凋亡特征,琼脂糖电泳可见间隔200bP的DNA梯形图谱,MTT线粒体功能降低。 he oxidative stress to induce apoptosis was studied in vitro in human peripheral blood lymphocytes (HPBLs). HPBLs were exposed in 400 μmol/L H 2O 2 6 8 hrs and the morphological changes of characteristic apoptosis were revealed by light and electron microscopic studies. Upon agarose gel elecrtophoresis the fragmented DNA of HPBLs treated with H 2O 2 for 8 hrs was demonstrated a ladder pattern of about 200 base pair increment manner which is typical of apoptosis. MTT assay demonstrated a significant decrease of the activity of mitochondrial dehydrogenase after treated with H 2O 2. The results indicated that H 2O 2 might induce apoptosis in peripheral blood lymphocytes.
