摘要 用天然胶原蛋白加交联剂制备多孔明胶微球,直径30~60μm,含微孔。将其作为荧光金载体用于垂体前叶神经纤维起源的研究获得较理想结果,并有以下优点:1.吸收示踪剂硬结后可按需要捣成直径60μm以上坚硬颗粒,使用方便;2.供示踪剂附着的表面积大,运载示踪剂量多;3.示踪剂从微球中缓慢释放;4.明胶化学性质稳定。 orous gelatin microsphere (PGM) is made of collagen by using cross linking technique. PGM had a diameter of 30 60μm. When tracing the nerve fibers of rat anterior pituitary, PGM was used as the carrier of fluoral gold (FG). Several advantages of PGM used as tracer carrier in nerve tracing study were confirmed. First, its rigid character makes it easy to manipulate; Second, it possesses a large area of surface on which a great amount of tracer can be carried; Third, after the FG bearing PGM stayed in vivo for 7 days, then the retrogradely labled hypothalamus neurons were fluorescently bright, so were the PGM, suggesting a gradual release of the tracer; Fourth, owing to its chemical stability, gelatin does not influence the activity of the tracers, neither the ability of the nerve to take up them.
