摘要
为了提高纤维素酶的作用效率,降低薯蓣皂苷元提取过程中纤维素带来的传质阻力,从而提高薯蓣皂苷元产率,在酶解的预处理过程中对糖液进行分离并添加β-葡萄糖苷酶分解体系中的纤维二糖.通过对不同预处理方式进行比较,考察了β-葡萄糖苷酶在纤维素降解过程中的作用,分析了纤维素酶的作用机制和抑制机理,认为葡萄糖和纤维二糖对纤维素酶的抑制作用不可忽略.利用实验结论对酶催化提取薯蓣皂苷元的工艺进行改进,使产率提高了29.3%.
To enhance the efficiency of cellulose bio-degradation and reduce the mass transfer resistance from cellulose consequently increase the extraction rate of diosgenin, glucose was separated and β-glycosidase was added in to decompose cellobiose in the biocatalysis pretreatment. The effect of β-glycosidase on cellulose degradation was studied by groups of experiments in different conditions of pretreatment. By analyzing the mechanisms of degradation and inhibition, cellulase's inhibition of glucose and cellobiose in different pretreatment modes was significant and the method to eliminate the inhibition was got. The techniques of extraction of diosgenin were optimized by separating the glucose and adding in β-glycosidase, the yield rate was increased by 29.3%.
