摘要 目的:优化薏苡仁生物转化条件及比较其转化前后甲醇总提取物对A549肺癌细胞株的细胞毒活性。方法:采用L16(45)正交表,以甲醇总提取物得率为指标进行正交优选;用MTT法测定和比较转化前后薏苡仁甲醇总提取物对A549细胞的细胞毒活性。结果:薏苡仁的最佳转化条件为:5%的转化物含量,转化时间为10 h,转化温度为60℃;通过转化能明显提高甲醇总提取物对A549肺癌细胞的细胞毒作用。结论:本研究对薏苡仁的进一步研究及其它中药的生物转化具有一定的参考价值。 Objective: To optimize biotransformation conditions for Semen Coicis and compare its antitumor activity of mechanolic extract of Semen Coicis through biotransformation . Methods: Biotransformation condition was optimized by L16 (4^5 ) orthogonal test with the yield of mechanolic extract of Semen Coicis as index. The cytotoxicity of mechanolic extract on A569 tumor cell was measured and compared by MTT assay. Results: The optimum condition was the following, culture of Rhizopus oryzae accounted for 5% , and it was transformed for 10 h at 60℃. Mechanolic extract of Semen Coicis showed higher antltumor activity through biotransformation. Conclusion: The research is meaningful to further study Semen Coicis and other traditional Chinese medicine's biotransformation.
