摘要
目的:建立以高效液相色谱法同时监测氯丙嗪、氯氮平、氯硝西泮、地西泮、阿普唑仑血药浓度的方法。方法:用乙酸乙酯提取血清中的药物,并进样分析,其中色谱柱为NovapakC18,流动相为甲醇-乙腈-水-三乙胺-冰醋酸(16∶36.5∶47.5∶0.03∶0.02),检测波长为254nm,流速为1.5mL.min-1,柱温为50℃,灵敏度为0.01AUFS。结果:血清中5种药物检测浓度均在0.03~100μg·mL-1范围内线性关系良好,方法回收率为88.6%~98.7%,日内和日间精密度小于5.0%。结论:本方法简便、快速、灵敏、准确、用血量少,可用于临床用药监护以及急性药物中毒快速定性和定量分析。
OBJECTIVE: To establish an HPLC method for simultaneous monitoring of the blood concentrations of five psychotropic drugs (chlorpromazine, clozapine, clonazepam, diazepam and alprazolam) in plasma.METHODS:Drugs in serum were extracted with ethyl acetate, and analyzed using Nova pak C18 as column, and methanol- acetonitrile- water- triethy- lamine -ice acetic acid(16 : 36.5 : 47.5 : 0.03 : 0.02) as mobile phase.The detection wavelength was at 254nm, flow rate at 1.5mL · min^-1, column temperature at 50℃ ,and sensitivity at 0.01AUFS. RESULTS :A good linear relationship was seen with the serum concentrations of the five drugs in the detective range of 0. 03 -100μg · mL^-1 .The recoveries of the five drugs ranged 88,6%-98.7% .The intra- day and inter- day relative standard deviation(RSD) was less than 5.0% .CONCLUSIONS: This method is convenient, rapid, sensitive and accurate, which requires use of less amount of blood. It can be used to monitor clinical drug use,and conduct fast qualitative and quantitative analyses of acute drug poisoning.
