摘要
目的探讨考马斯亮蓝法用于测定动物药材中可溶性蛋白质含量的稳定性研究。方法采用紫外分光光度法,以蟾蜍干体为原料,牛血清白蛋白为对照,在595nm处测定蛋白质与考马斯亮蓝结合复合物的吸光值。结果实验表明,吸光度和蛋白含量间具有良好的线性关系(r=0.996),样品在10min内测定稳定(RSD=5.0%)。结论本方法可靠、简单、快速。
Objective To study the stability of the Bradford method for determination of soluble protein in animal materials. Method The absorption of CBB G-250 combined with proteins was determined by the UV-Spectrophotometry method with the detection wavelength at 595 nm, and the dried bufo was selected as sample, BSA as control. Results The method showed a good linear relationship between absorption and protein content (r=0.996). The sample was stable with in 10 man (RSD=5.0%). Conclusion This method was reliable, simple and rapid. ;
