摘要 目的:研究过氧化质体增殖物激活受体(Peroxisome proliferator-activated receptor,PPAR)γ2基因Pro12Ala多态性是否与Ⅱ型糖尿病(IDDM)的易感性相关。方法:通过PCR-RFLP方法,对275例糖尿病患者和286例正常人进行了PPAR基因Pro12Ala多态性分析,比较各组间等位基因型及等位基因频率的分布。结果:研究对象中共见有2种基因型,Pro/Ala杂合子等位基因型,Pro/Pro纯合子等位基因型。IDDM患者基因型Pro/Ala、Pro/Pro频率分别为10%、90%,x2=3.39,P>0 05,而对照组Pro/Ala、Pro/Pro频率分别为5%、95%;IDDM患者等位基因Pro和Ala频率分别为95%、5%,x2=3.27,P>0 05,而对照组Pro和Ala频率分别为98%、2%。结论:糖尿病患者与对照组之间Pro12Ala多态性频率存在差异,但不显著。PPARγ2基因Pro12Ala多态性与Ⅱ型糖尿病的易感性可能无关。 Objective: To investigate whether Peroxisome proliferator - activated receptor (PPAR) γ2 gene Pro12Ala polymorphism is associated with the susceptibility to diabetes mellitus (DM). Methods: In this experiment, we analyze the PPARγ2 gene Pro12Ala polymorphism in 561 Chinese subjects including 275 patients with IDDM and 286 control subjects by the PCR - PFLP method. The genotype frequencies for Pro/Ala,Pro/Pro were 10% and 90% , respectively, in the DM group ,and 5% ,95% in the control subjects. Results: The Pro/Ala frequency is not significantly higher in the patients than in the control group (P 〉 0.05), and so is the Ala allele, suggesting that the Pro/Ala genotype maybe not associated with the susceptibility to IDDM in our study group. Conclusions : There maybe no correlation between PPARγ2 gene Pro12Ala polymorphism and susceptibility to IDDM.
