摘要
目的:体外观察全反式维甲酸(ATRA)对人源膀胱移行细胞癌BIU-87细胞的缝隙连接蛋白Cx26表达的影响。方法:分别用终浓度为0μmol/L、1μmol/L和10μmol/L的ATRA作用72h后免疫细胞化学法和半定量RT-PCR观察连接蛋白Cx26的表达情况及mRNA水平。结果:对照组细胞Cx26表达为阴性,mRNA水平低,几乎不表达,经过维甲酸刺激后表达显著增高,mRNA水平也显著提高。结论:ATRA可以促进Cx26基因转录,提高mRNA水平,增加Cx26连接蛋白的表达。
Objective:To investigate the regulative effect of all - trans retinoie aeid(ATRA) on the expression of gap junction connexin 26(Cx26) in human bladder cancer cell line BIU - 87 in vitro. Methods:BIU - 87 cells were exposed to ATRA at different concentrations (0μmol/L, 1μmol/L and 10μmol/L, respectively) for 72 h, and then levels of expression of Cx26 and its mRNA was determined with immunocytochemistry and semi - quantity RT - PCR. Results: In the control group, the expression of Cx26 was negative, its mRNA expressed scarcely and the quantity of mRNA was very low. After treatment with ATRA, both the expression of Cx26 and the level of mRNA increased. Conclusion:ATRA can increase the quantity of the Cx26 mRNA and enhance the expression of its protein.
