摘要
利用模拟移动床结合离子交换法从酶转化液中连续分离L-苯丙氨酸。在模拟移动床分离过程中,L-苯丙氨酸的吸附波稳定在区Ⅲ,吸附损失为零,单位质量树脂对L-苯丙氨酸的吸附量可达到125g/kg树脂,提高了树脂利用率:利用区Ⅰ的解吸尾流解吸杂质,L-天冬氨酸、蛋白等杂质的解吸波稳定在区Ⅱ,先于L-苯丙氨酸脱离模拟移动床分离系统;L-苯丙氨酸在区Ⅰ被集中解吸,解吸液中L-苯丙氨酸的浓度达35g/L。实验结果表明该法的L-苯丙氨酸解吸收率达97.6%,单位质量L-Phe的氨水(0.5mol/L)消耗率仅为38.37L/kg。
A new separation method of L-phenylalanine (L-Phe) from liquid of transformation was studied, and based on ion exchange and continuous separation by simulated moving bed (SMB) with standing wave design. The results showed that adsorption wave of L-Phe stays in zone Ⅲ without any adsorption loss, and the adsorptive capacity of L-Phe is 125g/kg resin, whereas the utilization of resin is improved. Desorption waves of asparagic acid and protein stay in zone Ⅱ by wake flow of the extract from zone Ⅰ and desorption wave of L-Phe stays in zone Ⅰ. The L-Phe concentration of elution is above 35g/L, while 97.6% L-Phe is obtained from desorption by simulated moving bed with ammonia consumption (0.5mol/L) rate of L-Phe only 38.37L/kg.
