摘要
以纯度≥99%的番茄红素晶体为研究对象,通过体外培养入宫颈癌细胞(Hela)、人黑色素瘤细胞(LiBr)进行抑制肿瘤能力试验。MTT法、细胞生长曲线测定、平板克隆形成试验结果显示,番茄红素对体外培养Hela、LiBr细胞有显著的抑制作用(p<0.01),培养96h后半数抑制浓度(IC_(50))分别为10.2μmol/L和10.8μmol/L.存在显著的剂量—效应、时间—效应关系。培养10d后Hela各处理孔未见细胞集落形成,LiBr各处理孔仅有少量细胞集落形成。培养48h后,两种细胞培养上清液中乳酸脱氢酶(LDH)水平显著升高,番茄红素使Hela、LiBr细胞膜损伤,提示番茄红素对Hela、LiBr细胞的抑制与直接杀伤有关。
Cultivating Hela and LiBr assays in vitro were used to inhibit tumor with about 99% purity lycopene crystals test samples. The results of MTT(methy lthiazolydiphenyl-tetrazolium bromide), cell growing curve and plate cloning showed that the lycopene crystals inhibition on Hela and LiBr was prominent p〈0.01. IC50 were 10.2μmol/L and 10μmol/L, respectively. The dose-effects relationship and time-effects relationship were prominent. Cell colony of Hela was not found after 10 d cultivation. And a few cell's colonies were found in both Libr and LDH cell fluids was increased prominently after 48h. The cell membranes of Hela and LiBr were damaged by lycopene crystals. It indicated that the inhibition on Hela and LiBr cells of lycopene has direct killing rerlationship.
