转录因子GATA—1基因在白血病中的表达

摘要 转录因子GATA家族在造血细胞的正常发育中起着重要的作用。利用聚合酶链反应(PCR)方法分析了116例各种白血病中红系特异转录因子GATA—1的表达情况。ANLL、CML、C-ALL和CLL中的表达率分别为43.75%、88.24%、14.29%和33.33%;3例T-ALL均不表达该基因。从80例ANLL中发现,MO、M1和M2亚型的GATA-1回表达率较高,而3例M3型及HL60细胞侏均不表达该基因。结果提示GATA—1基因在多种白血病中的异常表达情况,它在粒单系白血病细胞的持续表达现象,可能与在干细胞或粒单系祖细胞水平的异常增殖、分化和成熟障碍并致白血病有关。 Transcription factor GATA family play an important role in the normal developmental processof hematopoietic cells . Using polymerase chain reaction (PCR) method the expression of erythroid specific transcription factor GATA-- 1 gene in 116 cases of leukemin was analyses. Expression later of GATA-- 1 gene in ANLL, CML, C--ALL and CLL were 43.75%, 88.24%, 14.29%and 33.33%, respectively, but no expression round in 3 cases of T--ALL. Highly expression of GATA-- 1 in MO, M1 and M2 subtype of 80 cases with ANLL was found. Bat completely negative expression was seen in 3 cases with M3 and HL60 (M3 cell line). The data show the phenomenon of GATA-- 1 gene aberrant expression in leukemia and suggest that GATA-- 1 gene continuous expression in granulocytic and monocytic leukemia cells mas be relate to abnormal proliferation and a block in the nounal pattern of differentiation andmaturation at the level of hematopoietic stem cell or early myeloid progenitor that result in leukermia.
