摘要
目的:建立反相高效液相色谱法同时测定祁连獐牙菜中3种有效成分含量的方法。方法:采用ZORBAX SB-C18(250mm×4.6mm,5μm)色谱柱,流动相甲醇和水(含0.04%磷酸)的比例在0—20min内由20:80至30:70梯度洗脱,流速1ml/min,检测波长240nm,柱温35℃。结果:落干酸、獐牙菜苦苷、龙胆苦苷与其他成分达到基线分离。线性范围分别为0,05~0.625μg(r=0.9999)、0.0095~2.9μg(r=0.9998)和0.0486~2.56μg(r=0.9996),加标回收率分别为103%(3.9%)、98,2%(4.0%)、101%(3,5%)。结论该方法简便、快速,结果准确、可靠。
The objective of the research was to establish a quantitative method of simultaneously determination of loganic acid, swertiamarin and gentiopicroside in Swertia przewalskii with RP-HPLC. The samples were separated in the column of Zorbax SB-C18(250 mm × 4.6 mm,5μm) ,which eluted with methanol and water(0.04% phosphoric acid). The ration of methanol and water was increased from 20:80 to 30:70 in 20 min with a detected wavelength of 240 rim, the flowing rate of 1 ml/min,column temperature of 35 ℃. The result indicated three conpounds were isolated. The linear range of loganic acid, swertiamarin and gentiopicroside were 0.05 - 0.625 μg( r -- 0.999 9) ,0.009 5 - 2.9( r = 0.999 8)and 0.048 6 ~ 2.56 μg( r = 0.999 6), respectively. The average recoveries were 103%(3.9%),98.2%(4.0%) and 101%(3.5%),respectively.
