摘要
建立一种准确的测定全血铁、锌、镁的新方法.全血样经过硝酸-硫酸-高氯酸消化后,采用微量注射进样-火焰原子吸收法测定了Ⅱ型糖尿病患者全血中铁、锌、镁的含量.实验结果为3种元素的线性范围分别是5.0~25,0.1~1.0,0.5~4.0mg/L,相关系数r均在0.999以上,平均回收率分别为99.86%,94.88%,99.60%,相对标准偏差分别为1.40%,4.83%,1.63%,检出限分别为0.2107,0.0298,0.0146mg/L:用此方法测定104名Ⅱ型糖尿病患者的铁、锌、镁含量平均值分别为197.9±60.26,4.808±1.949.23.35±7.731mg/L,且均具有显著性差异(P〈0.001).实验证明该法具有样品需要量小、灵敏、准确、精密、快速、廉价等特点,适用于现代医学研究和临床检测.
A new and accurate assay method for the determination of iron, zinc and magnesium concentration in whole blood was establshed. First, whole blood samples were digested by nitric acid-sulfuric acid-perchloric acid. Then, iron, zinc and magnesium concentration was examined by FAAS with Micro Injection technique. The linearity of the three elements were 5.0 - 25, 0. 1 - 1.0, 0.5 - 4.0 mg/L, respectively, r 〉0. 999. The recoveries of whole method were 99.86%, 94.88 % and 99.60 % respectively. Relative standard deviations were 1.40 %, 4.83 % and 1.63 96 respectively. The detecting limits were 0. 210 7,0. 029 8, 0.014 6 mg/L respectively. The average value of iron, zinc and magnesium in whole blood of the Ⅱ Diabetes Mellitus were 197.9 ± 60.26,4. 808 ±1. 949,23.35 ± 7. 731 mg/L respectively . The content of three elements for the Ⅱ Diabetes Mellitus are lower than the healthy people and the difference is remarkable. The characteristic of this method were single sampling with small volume blood, the sensitivity, accuracy, precision, quickness and low-cost as well. This method is suitable to be used in clinical laboratories.
